First Minister Ian Paisley has said he believes Peter Robinson will become first minister on Thursday.
He said it would be an "evil thing" if anyone "so late in the day would now say no, we're stopping it".
There has been speculation Sinn Féin may not renominate Mr McGuinness when Mr Paisley stands down.
The party is widely acknowledged as being frustrated that the DUP is using its veto to block a range of issues, including the devolution of policing and justice.
Mr McGuinness said he hoped the outstanding issues could be resolved before Thursday.
"What we are dealing with is the need for all sides in this equation to recognise, 18 months on from the St Andrews Agreement, the absolute urgency required to see that agreement fulfilled," he said.
"Discussions are continuing as we speak. We will continue for as long as it is necessary to see the situation in a fashion that will convince people that there is an urgent desire to have the St Andrews Agreement fulfilled."

The DUP have been throwing their weight around since power sharing begun, SF are evidently aware that their electoral weight has subsided since the TUV ate up some of their vote in a local election.
The DUP refuse to honour the power sharing agreements which have understandably caused tension.
Do SF have a chance to grab more power in Stormont, this could be their best ever chance to become the largest party or close to it.
FF havent organised enough in this short space of time to be able to eat up SF's vote, so will they try to punish the DUP for not living up to the St.Andrews agreement?
Is it time SF stopped bending over for unionists and started using their own mandate to make life equally as difficult for them to show them that co-operation was a better option?

). I guess we'll find out tomorrow if this is much ado about nothing. I would say it'll be a popular move among the average northern nationalist voter if SF show they've the cojones to stand up to DUP intransigence on issues of importance to said nationalists.
